摘要
目的:建立甘草汤中总多糖的含量测定方法。方法:采用苯酚-硫酸法,以葡萄糖为对照品,在490 nm处以5%苯酚的用量、浓硫酸的用量、反应温度和反应时间为考察因素,优选最佳显色条件。结果:苯酚-硫酸法测定多糖的最佳显色条件为5%苯酚0.6 mL、浓硫酸4.5 mL反应温度25℃和反应时间30 min;葡萄糖质量在0.02~0.20 mg范围内与吸光度呈良好的线性关系,r=0.999 0;测得该方中总多糖平均含量为40.00 mg.g-1,RSD为1.23%。结论:采用苯酚-硫酸法测定该方中总多糖含量,快速准确,稳定可靠,能够作为该方总多糖含量测定方法。
Objective:To establish the content determination method of total polysaccharides in roasted liquorice decoction.Methods:Total polysaccharides were assayed by phenol-sulfuric acid method,referred to the grape sugar,and optimized the best color conditions,which regarded the dosage of 5% phenol and sulfuric acid,reaction temperature,and reaction time as observation factors.Results:The best color conditions of polysaccharides which were determinated by phenol-sulfuric acid method were 5% phenol 0.6 mL,sulfuric acid 4.5 mL,reaction temperature of 25 ℃ and reaction-time 30min;the quality of glucose at the range of 0.02-0.20 mg had a good linear relationship with absorbance,r=0.999 0;the average contents of total polysaccharides in the prescription were 40.00 mg·g-1 and RSD was1.23%.Conclusion:Phenol-sulfuric acid method used to determinate the content of total polysaccharide in the prescription is fast,accurate,stable and reliable,which can be regarded as content determination method for the total polysaccharide in roasted liquorice decoction.
