赝靛及其制剂中马卡因(Maackiain)的薄层扫描含量测定

Quantitative Determination of Maackiain in Radix Baptisiae Tinctoriae and Its Preparation

目的 :采用薄层色谱扫描法测定赝靛及其制剂施保利通口服液 (EsberitoxN)中指标成分马卡因 (Maackiain)的含量 ,为了考察不同色谱法定量测定的相关性 ,与高效液相色谱法测得的结果进行比较。方法 :样品经预处理后点于硅胶6 0F2 54 预制板上 ,以甲苯 丙酮 (8∶2 )为展开剂 ,于 30 6nm波长处进行吸收扫描 ,所得结果与HPLC测得的结果比较。结果 :此方法的平均回收率为 99.7% ,最低检测限为 0 .2 4μg ,符合中草药制剂的定量要求。 结论 :TLCS法与HPLC法比较 ,测定结果相关性良好。薄层扫描法和高效液相色谱法是互为补充的两种不同体系的色谱法 。

Objective:To determine Maackiain in Radix Baptisiae tinctoriae and its preparation Esberitox N Oral Liquid by TLC scanning. The results were compared with that got by HPLC to investigate the relativity of different chromato graphic methods. Methods:The sample solution was applied on a commercial silica gel 60F 254 precoated plate (Merck), developed with toluene acetone (8∶2) as a developing solvent system, and absorbed and scanned at 306nm of wavelenth. The results got by TLC scanning and HPLC were compared. Results:The average recovery was 99.7%. The limit of detection was 0.24μg, which conformed to the requirement of quantitative analysis of herbal medicinal. Conclusion:As being compared with HPLC method, the determination results got by means of two methods show a good correlation. TLC and HPLC are two kinds of different chromatographic systems which are complementary to each other. TLC scanning has the advantages of simplicity, fastness and flexible post chromatographic derivatization.
