摘要
通过分离纯化和酶学性质测定 ,确定耐热碱性磷酸酯酶 (TAPND2 7)为八聚体 ,最适反应温度为 6 5℃ .在 99℃保温 30min仍保留 49 4%的活性 ,在质量分数为 5 %的SDS和Tween2 0体系中 ,依然有 70 %以上的酶活性 ,并且有很宽的pH稳定范围 .这表明TAPND2 7适合在高温条件和高浓度去污剂的体系中应用 .
Through purification and characteristic determination, it was found that TAPND27 is a octamer with 65 ℃ as its optimal reaction temperature. It remains 49.4% enzymatic activity after incubation at 99 ℃ for 30 min while it remains above 70% enzymatic activity with 5% SDS or Tween20 in the reaction system. It is also stable in wide range pH. The results implied that TAPND27 is suitable for the application on the condition of high temperature and concentration of detergent.
