摘要
选用经初步筛选的 2 0个短序列引物 (10nt)和随机抽取的 2个随机长序列引物 ,对激光诱导产生的无核沙田柚 (处理组 )及未经激光处理的有核沙田柚 (对照组 )进行了RAPD(随机扩增多态性DNA)分析 .结果有 6个引物扩增出了共 2 0条多态性片段 ,从而表明在沙田柚的处理组与对照组之间 ,存在DNA分子水平上差异 ,也就是说在激光诱导沙田柚无核化的过程中 ,已经引起了沙田柚后代基因组的显著变异 ,且这种诱导所引起的性状改变是可以遗传的 ,从而为无性繁殖这种优良品种的可能性提供了理论上的依据 .
Twenty short arbitrary primers (10 nt) and two long ones (20 nt) were used in random amplified polymorphic DNA (RAPD) analysis for different strains of Shatian Pumelo. Among them, six primers could generate twenty polymorphic bands. With RAPD method, some genetic variations were shown between the Shatian Pumelo strains irradiated by laser and the wild strain. Further analysis of RAPD demonstrated that laser irradiation can induce the genetic variation of Shatian Pumelo.
