摘要
目的探讨复方蚂蚁酒对大鼠佐剂性关节炎的防治作用及其机制。方法选用60只SD大鼠,将其随机分为正常对照组、模型对照组、雷公藤多苷组和复方蚂蚁酒低、高剂量组(剂量分别为2.4、4.8 g/kg),每组各10只。各组给药3 d后,除正常对照组外,均采用弗氏完全佐剂足跖皮内注射复制佐剂性关节炎模型。于造模后第12天开始给药治疗,连续12 d。观察不同时间段的大鼠足趾肿胀率,检测大鼠血清一氧化氮(NO)、白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)的含量。结果与模型对照组和白酒对照组比较,雷公藤多苷组和复方蚂蚁酒低、高剂量组大鼠原发性和继发性足趾肿胀明显减轻,大鼠血清NO、IL-1β和TNF-α含量明显降低(P<0.05或P<0.01)。结论复方蚂蚁酒能减轻佐剂性关节炎大鼠的原发性和继发性足趾肿胀,其机制可能与其降低大鼠血清NO、IL-1β和TNF-α含量有关。
Objective To discuss the therapeutic effect and mechanisms of Compound Ant Wine on adjuvant arthritis in rats. Methods 60 SD rats were classified as normal control group, model control group, tripterygium glycoside group, low dose and high dose group of Compound Ant Wine with 2.4, 4.8 g/kg respectively, 10 in each group. All groups were used rats for adjuvant arthritis model induced by Complete Freund's adjuvant after 3 days of drug treatments except normal control group. The rats were treated with continuous 12 days after 12 days of model induced. The paw swelling rate of the rats was observed for different time periods, contents of rat serum nitric oxide (NO), interleukin-1 (IL-Iβ) and tumor necrosis factor-a (TNF-a content were measured. Results Compared with normal control group and wine control group, primary and secondary paw swelling of tripterygium glycoside group, low dose and high dose group of Compound Ant Wine were significantly attenuated, serum NO, IL-1β and TNF-a of the rats were significantly reduced. Conclusion Compound Ant Wine is capable of attenuating primary and secondary paw swelling of adjuvant arthritis in rats. The mechanism is related to reducing the contents of rat serum NO, IL-1 β and TNF-a.
