固定时间对口腔黏膜鳞状细胞癌组织染色的影响 被引量:3

The effects of fixation time on the staining for oral squamous cell carcinoma (OSCC) tissues

摘要 目的探讨固定时间对口腔黏膜鳞状细胞癌组织HE染色和免疫组织化学(immunohistochemistry,IHC)染色的影响。方法选取10例患者口腔黏膜鳞状细胞癌组织,每例各取4块,分别固定8h、24h、48h和1周,进行HE染色和高分子量细胞角蛋白(cytokeratin high molecular weight,CKH,克隆号34βE12)和广谱细胞角蛋白(cytokeratin pan,ckpan,克隆号AE1/AE3)IHC染色,观察细胞形态的优良、染色情况及IHC染色的阳性强度和阳性率。结果 HE染色和IHC染色结果均显示口腔黏膜鳞状细胞癌组织形态和染色、阳性强度和阳性率最佳为固定24h,8h为最差。固定1周者染色效果较48h差。结论本研究表明对口腔黏膜鳞状细胞癌组织,24h为最佳固定时间;组织固定不充分对HE染色和IHC染色的影响要大于固定时间过长的影响;固定时间越长,对HE染色和IHC染色效果影响越大。 Objective To investigate the effect of fixation time on HE staining and immunohistochemical (IHC) stain- ing of oral squamous cell carcinoma. Methods Ten cases of oral squamous cell carcinoma tissue were selected with 4 pieces in each case, they were respectively fixed for 8 h, 24 h, 48 h and 1 week. And they were conducted with HE staining and IHC staining with cytokeratin high molecular weight (CKH, clone: 34 [3 E12)and cytokeratin pan (CKpan, clone: AEI/AE3), the cell morphology, stain and the positive intensity and positive rate of IHC stain was observed. Results HE and IHC staining results showed that the tissue morphology, stain and the positive intensity and positive rate of oral squamous ceil carcinoma was best with fixation time for 24 h, and that was worst with fixation time for 8 h. The stain effect of fixation time for 1 week was worse than that for 48 h. Conclusion This study shows that 24 h is the best fixation time for oral squamous cell carcinoma, un-sufficient tissue fixation time has a greater effect on HE and I- HC stain than overlong fixation time. The longer fixation time, the greater effect on HE and IHC stain.
