摘要 目的研究Cripto-1在食管鳞癌组织中的表达情况及其与食管鳞癌发生、发展的关系。方法采用免疫组化SP法和原位杂交方法对60例食管鳞癌及60例正常食管黏膜组织进行Cripto-1蛋白和mRNA的检测。结果 (1)原位杂交结果显示,Cripto-1 mRNA在食管鳞癌组织中表达的阳性率为63.33%,显著高于正常食管黏膜组织中Cripto-1 mRNA的表达的阳性率(8.33%),两组之间Cripto-1 mRNA表达的差异具有统计学意义(P<0.05)。(2)免疫组织化学结果表明,Cripto-1蛋白在食管鳞癌组织中表达的阳性率为58.33%,显著高于正常食管黏膜组织中Cripto-1蛋白的表达的阳性率(6.67%),两组之间Cripto-1蛋白表达的差异具有统计学意义(P<0.05)。(3)对60例食管鳞癌患者的临床病理学参数进行统计分析,结果显示Cripto-1 mRNA和蛋白的表达均与食管鳞癌患者的年龄和性别无关(P>0.05),但与食管鳞癌的组织学分级、浸润深度、淋巴结转移和TNM分期具有显著的相关性(P<0.05)。(4)相关性分析结果表明,在60例食管鳞癌标本中,Cripto-1 mRNA阳性表达的38例患者中,Cripto-1蛋白阳性表达患者占31例,而Cripto-1 mRNA阴性表达的22例患者中,Cripto-1蛋白阴性表达占18例。Cripto-1 mRNA和蛋白在食管鳞癌组织中的表达呈正相关关系(γ=0.600,P=0.000)。结论食管鳞癌组织中Cripto-1 mRNA和蛋白均呈现高表达,表明Cripto-1高表达可能与食管鳞癌的发生和进展密切相关。 Objective To explore the correlation of Cripto-1 mRNA and protein expression with the occurrence and development of esophageal squamous cell carcinoma (ESCC). Methods SP immunohistochemical method and in situ hubridyzation method were used to detect the expression of Cripto-1 protein and mRNA in 60 cases of esophageal squamous cell carcinoma specimens and 60 cases of cases of corresponding normal esophageal epithelial tissues. Re- suits (1) The result of in situ hybridization demonstrated that positive ratio of Cripto-1 mRNA in ESCC tissues was 63.33%, which was significantly higher than those in normal esophageal epithelial tissue (8.33%), and there were sig- nificant differences in Cripto-1 mRNA expression between ESCC tissues and normal esophageal epithelial tissues (P 〈 0.05).(2) The result of immunohistochemistry showed that positive ratio of Cripto-I protein in ESCC tissues was 58.33%, which was significantly higher than those in normal esophageal epithelial tissue (6.67%), and there were sig- nificant differences in Cripto-I protein expression between ESCC tissues and normal esophageal epithelial tissues (P 〈 0.05). (3)For statistical analysis of clinicopathological features of 60 cases of the patients with ESCC, the results re- vealed that expressions of Cripto-1 mRNA and protein were not both related to age and gender of the patients with ESCC (P 〉 0.05), but closely associated with histological grade, invasion depth, lymph node meatstasis and TNM stag- ing (P 〈 0.05). (4)The results of correlation analysis demonstrated that the number of Cripto-I mRNA-positive was 38 in 60 cases of ESCC tissues, in which 31 cases displayed Cripto-1 protein-positive expression. Whereas, 22 cases of Cripto-1 mRNA-negative ESCC tissues appeared 18 cases of Cripto-1 protein-negative expression. Cripto-1 mRNA and protein expressions displayed positive correlation (γ=0.600, P = 0.000). Conclusion Cripto-I mRNA and protein both appear high expressions in ESCC tissues, indicating that Cripto-1 at high level may be closely associated with the development and progression of ESCC.
