摘要
采用氨基聚丙烯膜分别进行衍生引入氨苯基 (AP)和对苯二异硫氰酸基 (DITC)生成AP聚丙烯膜和DITC聚丙烯膜两种用于蛋白质固相序列测定的膜载体 .用上述膜载体偶联样品进行Edman降解、测序 ,结果表明 :蛋白质样品在上述两种膜载体的偶联率分别达到6 8 34%及 78 6 3% ,序列分析的重复收率分别达 90 6 9%及 91 4 7% .实验中两种膜载体表现出较好的机械与化学稳定性 ,且制备方法简单 ,成本较低 ,因而是固相蛋白质序列分析较好的载体材料 .
The preparation of derivatized amino polyp ropylene (APP) membrane by introducing aminophenyl group (AP) or isothiocyanate group (DITC) respectively for solid-phase protein sequencing is described. The avarage attachment yield of protein samples to the AP-APP and DITC-APP membran e was 68.34% and 78.63% respectively, the repetitive yield for Edman degrada tion of the AP-APP membrane and DITC-APP membrane was 90.69% and 91.47% respective ly. Both of the two kinds of the membrane showed good mechanical and chemical st ability during sequencing, and the advantage of easy to prepare with relativ ely low cost.
