肝癌肝动脉化疗栓塞治疗对缺氧诱导因子2α表达的影响及其临床意义 被引量:5

摘要 目的:探讨采用肝动脉化疗栓塞(transarterial chemoembolization,TACE)治疗的肝癌患者肝癌组织中缺氧诱导因子2α(hypoxia inducible factor 2alpha,HIF-2α)的表达及其临床意义。方法:采用30例TACE治疗后手术切除的肝癌患者(研究组)的肝癌组织标本以及30例直接手术切除的肝癌患者(对照组)的肝癌组织标本,采用实时荧光定量逆转录-聚合酶链反应(RT-PCR)法、蛋白质印迹法(Western blot)、免疫组织化学(IHC)方法检测肝癌组织中HIF-2α的表达水平并分析其临床意义。结果:研究组肝癌组织中HIF-2α的mRNA和蛋白水平显著高于对照组(P<0.05);肝癌组织中HIF-2α蛋白表达水平与肝癌患者的总体生存时间相关(95%可信区间为0.442~0.972,P<0.05),而与无瘤生存时间无关(P>0.05)。结论:TACE治疗可使肝癌组织中HIF-2α表达水平显著上升,其表达水平与恶性表型相关。 Objective:To investigate the expression of hypoxia inducible factor 2 alpha(HIF-2α) and its clinical significance in patients with hepatocellular carcinoma treated with transarterial chemoembolization(TACE).Methods:HIF-2α mRNA and protein levels were detected by quantitative real-time reverse transcription polymerase chain reaction and Western blot in the tissues of hepatocellular carcinoma from patients underwent TACE and operation(study group,n=30) and patients merely underwent operation(control group,n=30).Expression of HIF-2α was further studied using immunohistochemistry.Finally,the clinical significance of HIF-2α was analyzed.Results: Expression levels of HIF-2α mRNA and protein were significantly up-regulated in the study group compared with those in the control group(P0.05).The level of HIF-2α protein was significantly correlated with overall survival(95 % confidence interval: 0.442~0.972,P0.05),but not correlated with disease-free survival(P0.05) in patients with hepatocellular carcinoma.Conclusions: TACE treatment significantly increases HIF-2α expression level,and the level of HIF-2α is correlated with malignant phenotypes of hepatocellular carcinoma.
