基质金属蛋白酶-3与动脉粥样硬化

摘要 基质金属蛋白酶(matrix metalloproteinases,MMPs)是由23种(大鼠为24种)锌离子依赖的肽链内切酶组成的蛋白酶家族,1962年由Gross等[1]在研究蝌蚪尾巴变形时发现,因能降解胶原纤维而被称为间质胶原酶。MMPs能水解几乎所有的细胞外基质成分,参与了动脉粥样硬化(atherosclerosis,AS)等许多疾病的病理过程。基质金属蛋白酶-3(matrix metalloproteinase-3,MMP-3)属于MMPs家族中第3类即基质溶解素类,具有广泛的底物特异性,是MMPs的重要成员之一。AS是许多心脑血管疾病的病理基础。大量研究表明,MMP-3在AS的形成及发展过程中发挥了重要作用。
