胃溃疡组织中Smac和XIAP表达的研究 被引量:1

Expressions of Second Mitochondria-Derived Activator of Caspases and X-Linked Inhibitor of Apoptosis Protein in Gastric Ulcer Tissue

摘要 目的探讨促凋亡因子(Smac)和凋亡抑制蛋白(XIAP)在胃溃疡组织中的表达及意义。方法分别采用原位末端标记(TUNEL)法和免疫组织化学二步法检测40例胃溃疡患者溃疡愈合前后胃黏膜组织中细胞凋亡指数及Smac和XIAP的表达。结果胃溃疡愈合前组织中Smac阳性表达强度多为(++)~(+++),愈合后其阳性表达强度多为(+)~(++),溃疡愈合前Smac阳性表达明显强于愈合后(P<0.01);溃疡愈合前后组织中XIAP阳性表达强度比较差异无统计学意义(P>0.05)。溃疡愈合后细胞凋亡指数明显低于溃疡愈合前(P<0.01)。结论 Smac过表达可能促进胃黏膜上皮细胞凋亡而导致胃溃疡,并影响其愈合;Smac和XIAP蛋白可能参与了Hp感染性胃溃疡发生凋亡和增殖的信号通路。 Objective To explore the expressions of second mitochondria-derived activator of caspases (Smac) and X-linked inhibitor of apoptosis protein (XIAP) in gastric ulcer tissues and their roles. Methods The tissue specimens from dO patients with gastric ulcer were detected for apoptosis index by TdT-mediated dUTP nick-end labeling (TUNEL) and expressions of Smac and XIAP by immunohistochemistry(IHC) before and after treatment. Results The positive expression of Smac varied from positive ( + + ) to strongly positive ( + + + ) before treatment, and varied from positive ( + ) to positive ( + + ) after treatment,the positive expression of Smac before treatment was significantly higher than that after treatment(P 〈0.01 );XIAP before and after treatment showed no significant difference(P 〉0. 05). Apoptosis index after treatment was significantly lower than that before treatment (P 〈 0. 01 ). Conclusion Over-expression of Smac may promote apoptosis of gastric epithelial cells,lead to gastric ulcers and affect their healing. Smas and XIAP may involve in the signaling pathway of apoptosis and proliferation of Hp infective gastric ulcer.
