摘要
目的观察丹参酮ⅡA在体外对人皮肤恶性黑素瘤A875细胞的影响。方法体外培养A875细胞,0μg/ml、4μg/ml、8μg/ml、16μg/ml、32μg/ml丹参酮ⅡA作用A875细胞24 h、48 h,用MTT法检测丹参酮ⅡA对A875细胞增殖的影响;倒置显微镜观察细胞形态;Western blot法检测Caspase-3蛋白表达水平变化。结果丹参酮ⅡA有抑制A875细胞增殖作用,并且呈明显的浓度、时间相关性;随药物作用时间延长,Caspase-3蛋白表达量逐渐升高。结论丹参酮ⅡA促进Caspase-3高表达,诱导A875细胞凋亡,从而抑制细胞增殖。
Objective To study the effect of tanshinone ⅡA on human melanoma A875 cells in vitro. Methods A875 cells were cultured with different concentrations (0,4,8,16,32 μg/ml) of tanshinone ⅡA for 24 or 48 hours in vitro. The proliferation of A875 ceils was examined by MTT assay. The cellular morphology of A875 cells was observed by inverted microscope, and the changes of protein expression of Caspase-3 were detected with Western blot. Results A875 cells proliferation was significantly inhibited by tanshinone ⅡA in concentration and time-dependent manner. With the prolonged time of drug action,protein expression of Caspase-3 increased. Conclusion The hyper-expression of Caspase-3 protein might play an important role in promoting apoptosis of human melanoma A875 cells,thereby tanshinone Ⅱ can inhibit proliferation of human melanoma A875 cells.
