摘要
目的探讨不同粒径氯化钠粉对哮喘小鼠IL-4、IL-5水平及肺组织炎症指标的影响。方法 40只BALB/c小鼠随机分成对照组、哮喘模型组、超细氯化钠粉治疗组、大粒径氯化钠粉治疗组。采用OVA致敏和激发构建哮喘模型。对照组则利用PBS致敏、OVA激发。ELISA检测小鼠血清中IL-4、IL-5水平,肺泡灌洗液(BALF)观察白细胞计数及分类,制备肺组织石蜡切片。结果模型组BALF中白细胞总数及分类比率较对照组明显增高(P<0.05,P<0.01),超细氯化钠治疗组白细胞总数及分类比率较模型组明显降低(P<0.05);模型组IL-4、IL-5水平明显高于对照组(P<0.01),超细氯化钠粉治疗组和大粒径氯化钠治疗组IL-4、IL-5水平明显低于模型组(P<0.01,P<0.05)。结论超细氯化钠粉吸入治疗哮喘小鼠可下调其血清IL-4、IL-5水平,起到抑制气道炎症、治疗哮喘的作用。
Objective To investigate the influence of different particle size of sodium chloride powder inhalation on inflammatory markers in lung tissue and blood serum levels of IL-4,IL-5 of asthmatic mice.Methods 40 Balb/c mice were randomly divided into the control group,the model group,the therapy group with extra fine sodium chloride powder,and the therapy group with large particle size sodium chloride powder.Asthmatic mode was established via sensitizing and provoking ovalbumin,while the control group was sensitized with PBS and provoked with OVA.The levels of serum IL-4 and IL-5 were detected by ELISA.The white blood cells were detected in bronchoalveolar lavage fluid(BALF),and the lung tissues were prepared to paraffin section.Results The total white count and its classification ratio in BALF were obviously higher in the model group than in the control group(P0.01 and P0.05).The total white count and its classification ratio in BALF decreased more obviously in the therapy group with extra fine sodium chloride powder than in the model group(P0.05).The levels of serum IL-4 and IL-5 were the highest in the model group(P0.01 and P0.05).Conclusion Extra fine sodium chloride powder inhalation can lower the levels of serum IL-4 and IL-5,suppress airway inflammation,and cure asthma in the treatment of asthmatic mice.
