双酚A对雌性大鼠间脑组织Kisspeptin/GnIH基因表达的影响 被引量:1

摘要 目的探索环境雌激素双酚A(BPA)对脑组织新型神经生殖内分泌调节因子Kisspeptin/GnIH基因表达的影响。方法以去卵巢雌性大鼠为动物模型,腹腔给予不同剂量的雌二醇(E2)或BPA后,应用荧光定量PCR法检测BPA或E2对雌性大鼠间脑组织中Kisspeptin、GnIH及其各自受体基因表达的影响。结果 BPA各剂量组及E2处理组均显著降低大鼠间脑内Kisspeptin基因的表达量(P<0.05),而BPA高剂量组及E2处理组则明显升高间脑内Kisspeptin受体GPR54基因的表达量(P<0.01);大鼠间脑内GnIH基因的表达量在BPA低剂量处理组明显下降(P<0.01),而BPA高剂量及E2组无显著变化,各剂量组GnIH受体GPR147基因的表达量亦无显著性改变。结论 BPA能干扰生殖轴中的重要神经肽Kisspeptin及GnIH的表达,并且其干扰作用具有低剂量效应。 Objective To investigate the effects of different levels of BPA on the gene expression of Kisspeptin and GnIH. Methods The adult female SD rats were ovariectomized followed by giving different doses of oestradiol and BPA. The mRNA levels of Kisspeptin, GnIH and their receptors in diencephalon of ovariectomized female SD rats were measured by fluorescence quantitative PCR. Results Both oestradiol and BPA treatment significantly reduced the total mRNA expression of Kisspeptin in rat diencephalon ( P 〈 0, 05 ) ; whereas oestradiol and high dose BPA treatment increased the expression levels of GPR54 mRNA apparently (P 〈 0.01 ). The expression level of GnIH mRNA decreased apparently in the low dose BPA group (P 〈 0.01 ), whereas there were no changes in the E2 group and the high dose BPA group ; the ex- pression level of GPR147 was not influenced by E2 or either dose of BPA. Conclusion BPA can interfere the expression of Kisspeptin and GnlH which are the significant neuropeptides in reproduction axis, and the interference appears a low dose effect.
