血糖波动对糖尿病大鼠心肌细胞外基质表达的影响 被引量:1

摘要 【目的】探讨血糖波动对糖尿病大鼠心肌细胞外基质表达的影响。【方法】雄性Wistar大鼠30只,分为对照组(正常无糖尿病),波动性高血糖组(糖尿病大鼠定时给予降血糖处理),持续性高血糖组(糖尿病大鼠不给予降血糖处理),每组10只,均为常规喂养。饲养8周后,取血清测定纤维连接蛋白(FN)、层粘连蛋白(LN)和Ⅲ型胶原的表达并各组间进行比较。【结果】波动性高血糖糖尿病大鼠模型成功建立,波动性高血糖糖尿病大鼠心肌组织内FN、LN和Ⅲ型胶原的含量显著高于持续性高血糖组及对照组大鼠,其差异有统计学意义。【结论】糖尿病时血糖波动对心肌细胞外基质的影响,要强于平稳的高血糖状态。 [Objective] To explore the impact of blood glucose fluctuation on the expression of extracellular matrix in myocardium of diabetic rats. [Methods] Thirty Wistar male rats were divided into control group( n = 10), fluctuant high blood glucose group( n = 10) and persistent high blood glucose group( n = 10). The control group included normal rats without diabetes mellitus. Diabetic rats in the fluctuant high blood glucose group were given hypoglycemic drugs, while diabetic rats in persistent high blood glucose group were not given hypoglycemic drugs. All rats received conventional feed. After the feeding for 8 weeks, the levels of fibronectin(FN), laminin(LN) and type Ⅲ collagen in serum were measured and compared. [Results]The diabetic rat models with fluctuant high blood glucose were established successful- ly. The levels of FN, LN and type Ⅲ collagen in myocardiura of diabetic rats with fluctuant high blood glucose were markedly higher than those in rats of persistent high blood glucose group and control group, and there was significant difference( P 〈Q0.05). [Conclusion] The impact of blood glucose fluctuation on extracellular matrix in myocardium of di- abetes mellitus is stronger than that on extracellular matrix in myocardium at stable high blood glucose state.
