麦冬多糖对心肌缺血再灌注大鼠脂质过氧化反应的影响 被引量:3

Effect of Ophiopogonpolysaccharide on Lipid Peroxidation after Myocardial Ischemia Reperfusion in Rats

摘要 【目的】研究麦冬多糖(MDG-1)对心肌缺血再灌注大鼠脂质过氧化反应的影响。【方法】40只SD大鼠随机分为五组:假手术组、心肌缺血再灌注1h组、心肌缺血再灌注24h组、麦冬多糖治疗1h组、麦冬多糖治疗24h组。建立大鼠心肌缺血再灌注模型,采用试剂盒法检测各组大鼠血清中谷胱甘肽(GSH)、丙二醛(MDA)和超氧化物歧化酶(SOD)含量;采用荧光分光光度计检测各组大鼠心肌组织中活性氧(ROS)水平;采用real-timePCR和Westernblot方法检测大鼠心肌组织中iNOS和eNOS的表达水平。【结果】与假手术组比较,心肌缺血再灌注模型组大鼠血清中GSH和SOD含量下降,MDA含量有所增加,心肌组织中ROS水平、iNOS和eNOS表达也存在增加趋势,经麦冬多糖治疗后,GSH和SOD含量有所回升,而MDA、ROS水平呈现降低趋势,iNOS和eNOS水平也有所下降。【结论】麦冬多糖可缓解心肌缺血再灌注损伤,其作用机制可能与降低NOS水平、抑制脂质过氧化作用有关。 [Objective]To explore the effect of ophiopogonpolysaccharide(MDG-1) on lipid peroxidation after myocardial ischemia reperfusion in rats. [Methods] Forty SD rats were randomly divided into sham group, ischemia reperfusion lh group, ischemia reperfusion 24h group, MDG-1 treating lh group and MDG-1 treating 24h group. The rat models of myocardial ischemia reperfusion were made. GSH, MDA and SOD in serum o{ rats were tested by reagent kit. ROS in cardiac tissue was analyzed by fluorescence spectrophotome- ter. The expression o{ iNOS and eNOS in cardiac tissue were examined by Western blot and real-time PCR. [Results] Compared with the sham group, the contents of GSH and SOD in ischemia reperfusion group were decreased and the content of MDA was increased, otherwise the expression of ROS, iNOS and eNOS were higher than those in the sham group. After treatment with MDG-1, the contents of GSH and SOD were markedly elevated, whereas the expression o{ MDA, ROS, iNOS and eNOS were decreased. [Conclusion] MDG-1 can attenuate myocardial ischemic injury, which is associated with the decreasing of the expression of NOS and the inhibition of lipid'peroxidation.
