摘要
【目的】研究视网膜母细胞瘤(retinoblastoma,RB)组织和正常视网膜组织中claudin-1和snail的表达水平及其临床病理意义。【方法]42例RB组织和15例正常视网膜标本常规石蜡包埋切片,claudin-1和snail染色方法为EnvisionT”免疫组化法。【结果]claudin-1在RB组织中表达阳性率为50%,较正常视网膜组织(93.7%)明显降低(P〈O.01);snail在RB组织中表达阳性率为54.8%,较正常视网膜组织(6.7%)升高(Pd0.01);临床分期Ⅰ期、病理分化型、未侵犯视神经、生存期≥2年的RB病例claudin-1表达阳性率高于临床分期Ⅲ期、未分化型、侵犯视神经、生存期〈2年的病例,临床分期Ⅰ期、分化型、未侵犯视神经的RB病例;snail表达阳性率低于临床分期Ⅲ期、未分化型、侵犯视神经的病例,claudin-1在临床分期Ⅰ期与Ⅲ期的表达存在差异趋势但无统计学上的显著性(P〉O.05),snail的表达阳性率在无瘤生存期〈2年和≥2年组无统计学差异(P〉0.05),需进一步大样本实验的证实;RB组织中clau—din-1表达与snail表达明显不一致(Pd0.01)。【结论】claudin-1和snail表达水平可能是RB发生、进展、临床生物学行为及预后的标记物,claudin-1高表达和(或)snail低表达者可能预后较好。
[Objective] To explore the expression of claudin-1 and snail in retinoblastoma(RB) tissue and nor- mal retina tissue and the clinicopathological significance. [Methods] Paraffin sections of 42 RB tissues and 15 nor- mal retina tissues were made. Envision.9 immunohistochemistry method was used in the staining of claudin-1 and snail. [Results]The positive rate of claudin-1 expression in RB tissues was 50 % which was obviously lower than that in normal retina tissues(93.7 %). The positive rate of snail expression in RB tissues was 54.8% which was higher than that in normal retina tissues(6.7 %). The expression of claudin 1 in RB patients with clinical stage Ⅰ , differentiated type, uninvolved optic nerve and life span≥2 years was higher than those in RI3 patients with clinical stage Ⅲ , undifferentiated type, optic nerve involvement and life span〈2 years. The expression of snail in RB pa- tients with clinical stage Ⅰ , differentiated type, uninvolved optic nerve and life span≥2 years was lower than that in RB patients with clinical stage Ⅲ , undifferentiated type, optic nerve involvement and life span〈2 years. There was difference trend in claudin-1 expression between clinical stage Ⅰ and stage Ⅲ, but the difference was not signif- n icant in statistics. There was no significant difference in the positive rate of snail expression between the group with tumor-free survival time〈2 years and the group with tumor-free survival time≥2 years, but it should be con- firmed by further large-sample experimental study. The expressions of claudin-1 and snail in RB tissue were highly inconsistent. [Conclusion]The expression of claudin-1 and snail may be the markers of the pathogenesis, progres- sion, biological behavior and prognosis of RB. Patients with high expression of claudin-1 and(or) low expression of snail are more likely to have better prognosis.
