摘要
目的:研究氯化锂(LiCl)内源性激活小鼠触须垫区毛囊神经嵴细胞(neure crest cells,NCCs)Wnt/β-catenin信号通路对NCCs增殖的影响。方法:流式细胞技术检测不同浓度的LiCl作用不同时间后对NCCs细胞周期的影响;筛选出LiCl刺激NCCs的最佳浓度和时间,利用细胞免疫荧光及蛋白质免疫印迹法检测β-catenin和细胞周期蛋白CyclinD1、CylinE1的表达情况。结果:20mmol/L的LiCl作用24h,NCCs表现出最强的细胞增殖活性。LiCl刺激NCCs后,β-catenin在细胞内含量升高同时在核内积聚,CyclinD1和CylinE1表达量升高。结论:LiCl能够激活Wnt/β-catenin信号通路,促进细胞周期蛋白的表达,从而促进NCCs增殖。
Objective:To study the effects of LiC1 endogenous activation of Wnt/β-catenin signaling pathways of neure crest cells (NCCs) in mice tentacle mat area on NCCs proliferation. Methods : Adopt LiC1 in different concentrations for different time periods to investigate its effects on NCCs cell cycles by FCM. Select the best concentration and time period for LiC1 to stimulate NCCs Using immunofluorescence and Western blot to observe the expression of [3-catenin and the expression of cyclin CyclinD1 and CyclinE1. Resuits:The strongest proliferation activity of NCCs took place 24 h after 20 mmol/L LiC1 was used. After the stimulation, the level of β-catenin, and expression of CyclinD1 and CyclinE1 increased in NCCs. β-catenin accumulated in the cores. Conclusions:LiC1 can activate Wnt,/β-catenin signaling pathway, boost the expression of cyclin, and promote NCCs proliferation.
