摘要
目的探讨核转录因子NF-κB及炎症相关的CXC趋化因子IL-8在人子宫颈癌细胞株Siha、CaSki中的表达及意义。方法选择两种宫颈癌细胞(未转移的宫颈鳞癌细胞株Siha、宫颈癌肠转移细胞株CaSki细胞)采用细胞免疫化学SV两步法检测两种宫颈癌细胞中NF-κB及IL-8的表达情况;采用酶联免疫吸附试验ELISA法检测两种宫颈癌细胞上清液中IL-8的表达情况,同法检测两种宫颈癌细胞的蛋白提取液中NF-κB的表达情况。结果 NF-κB及IL-8在Siha、CaSki两种宫颈癌细胞中均高表达,且两者在肠转移的CaSki细胞中表达明显高于未转移的宫颈癌细胞Siha细胞,差异有统计学意义(P<0.05)。结论 NF-κB及IL-8在宫颈癌细胞株CaSki中的高表达与宫颈癌的转移及发展密切相关。
Objective To survey the expression and significance of transcription factor NF-κB and the chemokine IL-8 in human Siha,CaSki cervical carcinoma cell strains.Methods The SV two-step Immunohistochemistry Assay Kit were used to detected the expression of NF-κB and IL-8 in the cells and their protein extracts of non-metastasized cervical squamous cell carcinoma strain Siha and metastasized cell strain CaSki.Enzyme-linked immunosorbent assay(ELISA) was used to detect the expression of IL-8.Results Both NF-κB and IL-8 were highly expressed in cervical carcinoma cells,and their expression in the intestine-metastasized CaSki cells was higher as compared with the non-metastasized cervical carcinoma cells Siha,the difference was statistically significant(P0.05).Conclusion The high expression of NF-κB and IL-8 in cervical carcinoma cell strain CaSki is closely related to the metastasis and development of cervical carcinoma.
