摘要 观察蛋氨酸脑啡肽(MENK)对巨噬细胞-粒细胞集落刺激因子(GM-CSF)诱导的小鼠骨髓来源树突状细胞TLR-4和TNF-α表达的影响,探讨蛋氨酸脑啡肽对小鼠骨髓来源树突细胞免疫功能的调节机制。体外制备小鼠骨髓细胞用GM-CSF协同MENK诱导培养7 d后用RT-PCR法检测TLR-4和TNF-α的mRNA表达,用Western-Blot和ELISA法分别检测TLR-4和TNF-α蛋白的表达。结果表明,MENK协同诱导组TNF-α和TLR-4表达高于正常对照组(P<0.01)和GM-CSF诱导组(P<0.05)。显示MENK可以促进树突状细胞TLR-4和TNF-α的表达。 To observe the influence of Methionine enkephalin (MENK)on the expression of toll like receptor 4(TLR-4) and tumor necrosis factor-a(TNF-a) of DCs derived from murine bone marrow progenitors (BM-DCs)induced by MENK and to investigate the immunity regulatory mechanism of MENK on DCs. The BM-DCs were prepared and cultured in vitro, induced by GM-CSF and MENK. The mRNA expression of TLR-4 and TNF-a were detected by RT-PCR.The protein expression of TLR-4 and TNF-a were detected by ELISA and Western-Blot technique respectively after seven days. The expression of TLR-4 and TNF-a in MENK group were both increased compared to normal control group (P〈0.01) and GM-CSF group (P〈0.05). This suggested that MENK is probably to promote the expression of TLR-4 and TNF-a from gene and protein level for DCs.
