摘要
对红豆草、苜蓿、百脉根和鹰嘴紫云英的花药、茎尖和茎段进行试管培养,诱导花药愈伤组织培养基以B_5、N_6、MS分别附加2、4—D_(0.05-2)、KT_(0.5-3)、6—BA_(0.5-1.5),LH300,蔗糖3%、琼脂0.6—0.7%,配成12种培养基。红豆草、苜蓿、百脉根和紫云英的诱导出愈率分别为8.94%,10.88%,23.25%和8.07%。其中红豆草和苜蓿以B_5最好,百脉根和紫云英较适宜N_6。用-2—3℃低温处理,24—30小时红豆草可提高出愈率4.48倍。分化培养基以N_6和B_5附加2.4-D_(0.05-0.5),KT_(1-3)、IAA_(0.5)、NAA_(0.5-2)、蔗糖3%、琼脂0.5%配3种培养基。以N_6和B_5附加2.4—D_(0.05)、KT_3、NAA_2为最好。还可以一次培养成苗。其次是以N_6和B_5附加IAA_(0.5)。pH值为5.8—6,并对形成愈伤组织及其分化过程、培养方法、条件和影响因素及快速繁殖方法进行了研究。这项研究为豆科牧草育种新技术的研究作了有益的探索。
Anther, shoot tip and section of sainfoin, Iucerne, lotus, and mikvetch were cultured on B5.B6 or MS medium containing2.4-D0.05-2mg/1, KT0.5-3mg/1,6-BA0.5-1.5mg/1, LH300,sucrose3%, and agar0.6-0.7%, respectively. The rate of callus appearing for sainfoin,lucerne louts and milkevetch is 8.94%, 10.88%, 23.25% and 8.07% respectively, of the 12 media tested B5 is the best one for lucerne and sainfoin, while N6 is most suitable for lotus and milkvetch. Callus appearing rate of sainfoin is increased by 4.48 times after the culture treated under -2—3C for is 24-30h. The callus can grow into seedlings directly on some of media tested. They are N6 or B5 containing 2.4-DO.05mg/1, KT3mg/1, and NAA2mg/1, N6 or B5 containing IAA0.5mg/1, and PH adjusted to 5.8-6.0.
