小G蛋白Ran对胰腺癌细胞增殖和凋亡的调控 被引量:1

Regulation of Ran GTPase Signaling Pathway on Cell Growth and Apoptosis in Pancreatic Cancer Cells

摘要 目的:探讨Ras超家族的小G蛋白Ran GTPase在胰腺癌组织及细胞中的表达情况,及其对胰腺癌细胞PANC-1增殖和凋亡的影响。方法:选取胰腺癌石蜡标本及相应正常组织标本各27例进行免疫组织化学染色。用LipofectamineTM2000转染Ran干扰RNA(small interference RNA,siRNA)进入胰腺癌细胞系PANC-1干扰其表达,之后利用Western blot观察Ran的表达情况。运用MTT及流式细胞术分别检测各实验组细胞的增殖和凋亡情况。结果:免疫组化染色显示在胰腺癌组织中Ran的表达阳性率及得分均明显高于癌旁正常组织(P<0.05)。将干扰Ran siRNA转染进入细胞系PANC-1中,利用Western blot发现Ran的表达明显下调。并且通过MTT实验发现在胰腺癌细胞系PANC-1中下调Ran表达能明显抑制该细胞生长(P<0.05),并使PANC-1细胞凋亡显著增多(P<0.05)。结论:小G蛋白Ran GTPase在胰腺癌组织及细胞系中高表达,且Ran可以促进胰腺癌细胞PANC-1的增殖,抑制其凋亡。 Objective: To observe the expression of the Ran GTPase in the pancreatic cancer tissues and cells,and the effect of the Ran GTPase on cell growth and apoptosis in PANC-1 pancreatic cancer cells.Methods: A total of 27 paraffin-embedded specimens of primary pancreatic cancer and matched adjacent non-cancerous tissues for Immunohistochemistry.Ran expression was inhibited by small interference RNA,the change of protein level in PANC-1 cell was measured by Western blot.We ues the MTT and flow cytometry(FCM) to detecte the growth rate and apoptosis of PANC-1 cells.Result: The expression level of Ran in pancreatic cancer tissues was sig-nificantly higher than that in matched non-cancerous tissues(P0.05).After inhibiting Ran expression,the growth of PANC-1 cells was inhibited(P0.05).However,the apoptosis of PANC-1 cells was increased(P0.05).Conclusion: Ran was overexpressed in pancreatic cancer tissues compared with non-cancerous tissues,which was determined by immunohistochemical staining.In PANC-1 pancreatic cancer cells,Ran promotes cell growth and inhibits apoptosis.
