摘要 目的 :研究cAMP、cGMP在心肌细胞碱性成纤维细胞生长因子 (bFGF)基因表达调节中的作用。方法 :分离培养大鼠心肌细胞 ,加入 8-溴 -cAMP和硝普钠 ,升高细胞内cAMP、cGMP浓度 ,用原位杂交及免疫细胞化学的方法观察细胞内bFGF的表达 ,图象分析检测bFGF表达量的变化。结果 :在培养中加 8-溴 -cAMP的心肌细胞 ,2 4h时bFGFmRNA及蛋白表达均明显高于对照组 ,48h、72h仍然较强表达 (P <0 0 5 )。而加硝普钠的心肌细胞 ,bFGF表达明显低于对照组 ,以 48h最为显著 (P <0 0 1)。结论 :cAMP。 AIM: To investigate the role of cAMP and cGMP in the regulation of bFGF gene expression of rat cardiomyocytes. METHODS: 8-Bromo-cAMP and sodium nitroprusside were added into the media to raise the concentrations of cAMP and cGMP of cultured cardiomyocytes, in situ hybridization and immunohistochemistry methods were used to identify the mRNA and protein expression of bFGF. The amount of bFGF mRNA and protein expression were detected by computor imaging analysis system MIAS exe. RESULTS: At 24 h after adding 8-Bromo-cAMP(0 1 mmol/L), bFGF mRNA and protein expression elevated significantly , at 48 h and 72 h it still expressed higher than that of control group( P< 0.05). But after adding sodium nitroprusside (1 mmol/L),bFGF expression decreased significantly,and especially at 48 h. CONCLUSION; The regulation of bFGF expression of cardiomyocytes is related to cAMP signal pathway.
