模拟高原低氧对p53及其靶基因表达的调节

摘要 目的:初步探讨模拟高原低氧对高原动物和平原动物p53及其靶基因的调节。方法:高原鼢鼠和根田鼠暴露于7 km(8.0%O2,41.02 kPa)低氧6 h,SD大鼠分别暴露于2 km(16.0%O2,79.97 kPa),5 km(10.8%O2,54.02 kPa)和7 km(8.0%O2,41.02 kPa)低氧8 h,采用实时荧光定量PCR和Western blot研究低氧对三种动物肝脏p53及其靶基因的调节。结果:低氧下调高原动物肝脏p53mRNA和蛋白的表达,但上调平原动物p53mRNA和蛋白的表达;中度和严重低氧对平原动物肝细胞凋亡靶基因bax和IGFBP3有抑制作用,而对细胞周期停滞靶基因p21有激活作用。结论:高原动物和平原动物肝细胞p53及其靶基因在低氧应激时具有不同的表达模式。
