rhAng-1对局灶性脑缺血/再灌注大鼠血脑屏障血管内皮细胞间连接的影响 被引量:2

The influence of rhAng-1 on endothelial junctions of BBB after focal cerebral ischemia/reperfusion in rats

摘要 目的:研究重组人血管生成素1(rhAng-1)对局灶性脑缺血/再灌注大鼠血脑屏障通透性的影响及脑微血管内皮细胞间连接蛋白表达的变化。方法:大鼠随机分8组(n=10):①手术组;②缺血组;③~⑤生理盐水+缺血/再灌注12 h,48 h,7 d组;⑥~⑧rhAng-1+缺血/再灌注12 h,48 h,7 d组。用伊文思兰渗透性实验检测血脑屏障通透性;TTC法检测脑梗死面积;按Zea Longer评分标准对大鼠神经功能损伤进行评分;用免疫组化、RT-PCR和West-ern blot法检测occludin、ZO-1和VE-cadherin的表达。结果:rhAng-1可明显降低局灶性脑缺血/再灌注大鼠血脑屏障通透性,降低脑梗死面积;rhAng-1可使细胞间连接蛋白occludin、ZO-1和VE-cadherin的表达增加。结论:rhAng-1通过上调脑微血管内皮细胞间连接蛋白occludin、ZO-1和VE-cadherin的表达降低局灶性脑缺血/再灌注大鼠血脑屏障通透性。 Objedive: This study was pedormed to determine whether recombinant human angiopoiefin-1 (rhAng-1) decreases the permeability of the blood-brain barrier (BBB) in focal cerebral ischemia and reperfusion rats, whether RhAng-1 opens the BBB by affecting tight junction associated proteins zonula occludin- 1 ( ZO- 1 ), occludin and adherens junction protein vascular endothelial (VE)-cadherin. Meflmds: The rats were divided into eight groups randomly( n = 10) : sham-opemted group; (2)ischemia group; (3) - (5) ischemia/reperfnsion (middle cerebral arte occlusion and reperfnsion (MCAO/R) 12 h, 48 h, and 7 days) and 0.9% saline groups; ischemia/repeffusion (MCAO/R 12 h, 48 h, and 7 days) and rhAng-1 groups. The BBB permeability was assessed by Evans blue extravasation. The messenger RNA and imin expressions of ZO-1, occludin, and VE-cadhefin were determined by reverse transcfiption-polymerase chain reaction, West- ern blot, and inunondechemistry assays. Results: BBB pemieility and the brain infarct volume were significantly decreased after dutlng-1 injection. The expressions of ZO-1, oecludin, and VE-cadherin were increased after rhAng-1 injection. Conclusion: rhAng-1 may decrease the penneadty of BBB in MCAO/R rats by upgulation of ZO-1, eccludin, and VE-cadherin.
