摘要
目的:观察去势大鼠整合素表达水平,揭示补肾中药治疗骨质疏松症的作用机理。方法:10月龄雌性Wistar大鼠50只,随机分为5组:福善美组(FSM)、金刚健骨片组(JGJG)、骨松宝颗粒组(GSB)、模型组(MODEL)、假切组(SHAM)。假切组切除一段肠系膜,其余组均行卵巢摘除手术。饲养13周模型建立后,治疗组分别以相应药物连续给药13周,模型组及假切组以生理盐水替代。治疗结束后处死动物取出骨组织,进行骨密度测定、并利用免疫组化法测定整合素αⅤβ1及αⅤβ6表达水平。结果:骨密度测定结果显示,FSM组、JGJG组的股骨密度、整合素αⅤβ6均较MODEL组有明显提高,差异有统计学意义(P<0.05);FSM组、JGJG组及GSB组整合素αⅤβ1均明显低于MODEL组,差异有统计学意义(P<0.05)。结论:金刚健骨片能显著提高去势大鼠的股骨密度、整合素αⅤβ6的表达水平,并可降低整合素αⅤβ1的阳性表达率,从而干预成骨-破骨细胞偶联,达到治疗目的。
Methods:Fifty female wistar rats (10-months-old) were randomly divided into 5 groups: Fosamax group (FSM), Jingang Jiangu Pill group (JGJG), Gu Songbao Granule group (GSB), Model group (MODEL), and Sham group. A segment of mesenteric was cut in Sham group, the others groups were treated with ovariectomy. The rats were raised for 13 weeks to established model. They were treated with corresponding drugs for another 13 weeks, excluding model group and the sham --cut group which replaced with saline. BMD and the expression of integrin αvβ1 and αvβ6 were measured in each group after the treatment. Results: The BMD and the expression of integrin αvβ6 of FSM group and JGJG group were increased obviously compared with MODEL group (P〈0.05). But the expression of integrin αvβ11 in FSM group, JGJG group, and GSB group were significantly decreased compared with MODEL group (P〈0.05). Conclusion:JGJG pill can obviously improve BMD and the level of integrin αvβ6 in ovariectomy rats and reduce the expression rate of integrin αvβ1, to intervene the coupling of osteoblasts and osteoclasts for therapeutic purposes.
