摘要 目的研究模式识别分子mindin在体外HCV感染细胞系的表达状况。方法在体外将全长HCV嵌合体基因组电转染到Huh-7和Huh-7.5细胞中,用间接免疫荧光观察转染48h后HCV核心蛋白表达,用Western印迹方法检测转染72h后细胞内mindin蛋白水平的变化。结果 HCV核心蛋白在转染细胞中表达,在未转染的细胞中不表达;与未转染细胞相比,转染72h后细胞内mindin蛋白水平降低。结论 HCV感染下调肝细胞mindin表达,mindin可能参与HCV致病过程。 Objective To study the expression of pattern recognition molecule mindin in Huh7 and Huh7.5 cells harboring in vitro transfeeted full-length hepatitis C virus (HCV) RNA. Methods Plasmid J6/JFH encoding the full- length HCV chimeric genome was transfected to HCV in vitro and the RNA was transfected into Huh7 and Huh7.5 cells by electroporation. Indirect immunofluorescence was used to detect the expression of HCV protein. Western blot was used to detect the expression of mindin. Results Indirect immunofluorescence showed high expression of HCV core protein in the transfected cells. Western blot showed down-regulated expression of mindin in the transfected cells. Conclusion HCV down-regulates mindin in Huh7 and HuhT. 5 cells. Mindin may play a role in the pathogenesis of HCV.
