摘要
目的证实体外培养的大鼠膀胱上皮细胞是否可以通过MAPK及STAT信号途径调节脂多糖诱导的炎症反应。方法分离幼龄SD大鼠膀胱粘膜,培养上皮细胞,分为四组:对照组、脂多糖组、PD98059+脂多糖组及AG490+脂多糖组,采用Real time-PCR及Western blot技术检测ERK及Stat3表达量的变化。结果脂多糖可显著激活膀胱上皮细胞中ERK及Stat3的表达,PD98059及AG490能够部分减低脂多糖刺激后两种基因的相对表达量。结论 MAPK/ERK及STAT信号通路可能参与脂多糖诱导的大鼠膀胱上皮细胞的炎症反应。
Objective To determine the regulation of MAPK and STAT signaling pathways in cultured bladder epithelial cell of SD rat induced by lipopolysaccharide. Methods Cultured infant rat bladder epithelial cells were separated from rat bladder mucosa and divided into control group, lipopolysaccharide group, PD98059+lipopolysaccharide group and AG490+lipopolysaccharide group respectively. Real time-PCR and Western blot were used to determine the expression of ERK and Stat3. Results The expression levels of ERK and Star3 in the bladder epithelial ceils were increased significantly in lipopolysaccharide rats than in the controls, and PD98059 and AG490 could partly reduce the expression levels of ERK and Stat3. Conclusion MAPK/ERK and STAT signaling pathways might be involved in the inflammatory process of bladder epithelial cells induced by lipopolysaccharide.
