摘要
目的:研究高三尖杉酯碱(HHT)对人胃癌SGC-7901细胞诱导凋亡的作用,并探讨其可能机制。方法:常规培养人胃癌SGC-7901细胞,根据HHT浓度分为空白组和HHT小、中、大剂量组(剂量依次为20、80、120μg.ml-1)。采用MTT法观察HHT对SGC-7901细胞生长的抑制作用;用流式细胞术测定SGC-7901细胞周期的变化。结果:MTT比色法显示各浓度药物作用24、48和72 h均能显著抑制SGC-7901细胞的增殖(P<0.05);随着药物浓度的升高,SGC-7901细胞中S期细胞比例逐渐降低。结论:HHT能诱导胃癌SGC-7901细胞发生凋亡,其机制值得进一步研究。
Objective: To study the apoptosis of human gastric cancer SGC-7901 cells induced by homoharringtonine, and to explore its possible mechanism. Methods: Human gastric cancer SGC-7901 cells were routinely cultured, and were divided into blank group and a low, middle and high- dose homoharringtonine groups of 20,80 and 120 μg·ml-1 respectively. MTT colorimetry was used to observe the growth inhibition of SGC-7901 cells caused by harringtonine. SGC-7901 cell cycles were measured by flow cytometry. Results: MTT colorimetry showed that various levels of concentration of homoharringtonine at 24,48 and 72 h could significantly inhibit the proliferation of SGC-7901 cells(P〈0.05).The proportion of cells in S phase of SGC-7901 cells gradually reduced along with the increase of drug concentration. Conclusion: Homoharringtonine induced cell apoptosis of SGC-7901 gastric cancer cells, and its mechanism is worth further study.
