摘要
目的通过观察卵巢早衰POF患者外周血CD4+CD25+调节性T细胞Treg及干扰素-γ(IFN-γ)、转化生长因子-β(TGF-β)的变化,探讨POF的免疫学发病机制。方法收集2011年12月-2012年9月就诊的POF患者17例,卵巢储备功能减退DOR患者11例,以及生殖中心健康育龄女性16例,流式细胞仪定量检测外周血Treg数量,Elisa方法检测血清IFN-γ、TGF-β的水平,并以FSH/LH评价卵巢储备功能,进行相关性分析。结果与对照组相比,POF组和DOR组IFN-γ水平增高P<0.01、TGF-β水平降低P<0.01,POF患者及DOR患者Treg比例降低P<0.01,IFN-γ的增高与卵巢储备功能的下降呈显著正相关r=0.70,P<0.01。结论 Treg和IFN-γ、TGF-β水平与卵巢早衰密切相关,IFN-γ对评估卵巢储备功能、预测卵巢早衰具有参考价值。
Objective To investigate the autoimmune mechanism of premature ovarian failure(POF) by measuring the percentage of Treg and the concentration of serum interferon(IFN)-γ and transforming growth factor(TGF)-β1 in patients with POF.Methods Between December 2011 and September 2012,17 POF patients,eleven diminished ovarian reserve(DOR) patients from the obstetrics and gynecology clinic,and sixteen volunteers from the Reproduction Center were enrolled in the study.Flow-cytometry analysis of Treg was performed on fresh peripheral blood;serum IFN-γ and TGF-β1 level in POF and DOR patients and normal controls were detected by Elisa,and the correlation between cytokines and FSH/LH was analyzed.Results Compared with the control group,serum IFN-γ level was obviously higher(P 0.01) in POF and DOR group,while the serum TGF-β1 level was significantly lower(P 0.01).The percentage of Tregs in POF and DOR patients were obviously lower(P 0.01).A positive correlation existed between IFN-γ and DOR(r=0.70,P 0.01).Conclusions IFN-γ,TGF-β and Treg play important roles in autoimmune POF.IFN-γ can be regarded as an important symbol for evaluating ovarian reserve and predicting POF.
