结合响应面分析法优化复方金钱草中君药的半仿生提取工艺 被引量:9

摘要 目的结合响应面分析法,研究复方金钱草中君药的半仿生提取工艺最佳条件。方法利用响应面分析法,测定复方金钱草各半仿生提取液中的总黄酮、总糖质量分数和干浸膏质量,并进行综合比较,以综合评分为响应值作响应面和等高线,分析提取工艺条件。结果复方金钱草中广金钱草、路路通、车前子3种君药半仿生提取最佳条件为:温度100℃、料液比1 g∶35 mL,提取3次,3次提取液pH值分别为4.0、7.0和8.5,总提取时间为2.5 h。在此条件下,总黄酮质量分数为9.91 mg/g,总糖质量分数为62.38 mg/g,浸膏得率为13.86%,综合评分为467.4。结论复方金钱草中君药提取采用半仿生提取法优于水煎煮法。响应面分析法结合半仿生提取法可应用于复方金钱草的提取工艺优化,但对因素水平的变化范围和试验数据要求较为严格。 Objective To study on optimum conditions of semi-bionic extraction of the monarch drugs of Fufang Jinqiancao based on response surface analysis method.Methods The total flavonoids,total saccharides and dry extract with SEB theory were determined and compared by response surface analysis to optimize the extract conditions with the comprehensive grade as response value and contour line.Results The best semi-bionic extract conditions of the three monarch drugs of Fufang Jinqiancao were as follows:100 ℃ temperature,1 g∶35 mL solid-liquid ratio,extracting for three times,at pH4.0,7.0,8.5 for each solutions and total time for 2.5 h.The content for total flavonoids was 9.91,62.38 mg/g for saccharides,the yield of extract was 13.86% and the comprehensive score was 467.4.Conclusion The semi-bionic extraction of the three monarch drugs is better than the water decocting method.Response surface methodology combined with semi-bionic extraction can be applied to the extraction optimization of Fufang Jinqiancao,up to more exact demands for the variaion of influence factors and the experimental data.
