舒洛地特对慢性环孢素肾病大鼠的保护作用 被引量:1

摘要 目的观察舒洛地特对慢性环孢素肾病大鼠(CCN)的保护作用。方法 36只Wistar大鼠随机分为正常对照组、模型组、贝那普利阳性对照组、三组舒洛地特干预组,观察各组大鼠体质量、血尿素氮和肌酐的变化;实验末取肾组织行HE、PAS及Masson染色观察病理变化,分析各组小动脉透明样变性、肾间质纤维化情况。电镜观察肾组织超微结构变化。结果模型组大鼠体质量下降,血尿素氮、肌酐明显升高。4周后所有用药组体质量均高于模型组,尿素氮、肌酐水平均低于模型组;模型组大鼠可见典型CCN改变,各用药组大鼠的CCN病变明显减轻;模型组大鼠肾小球、肾小管基底膜增厚,细胞核固缩,间质见较多胶原纤维,各用药组病变较轻。结论舒洛地特干预用药可减轻环孢霉素肾毒性,早期干预效果更好,舒洛地特预防环孢霉素A肾毒性优于贝那普利。 Objective To observe the effect of sulodexide on the chronic cyclosporine nephropathy(CCN) in rats.Methods Wistar rats were randomly divided into 6 groups,including the normal control group,the model group,benazepril group,and three sulodexide groups with different dosages.Rat's body weight was checked,and blood was collected to measure the levels of blood urea nitrogen(BUN) and serum creatinine(Scr).Kidney tissues were collected and stained with HE,PAS and Masson.Pathological changes,hyaline degeneration of small arteries and renal interstitial fibrosis were assessed.The ultrastructural changes of kidney tissues were also observed by electron microscopy.Results After CsA administration,the weight of rats decreased and the levels of serum BUN and Scr elevated in the model group,but which was arrested in benazepril group and all sulodexide groups.Compared with the normal control group,CCN was found in the kidney tissues in the model group,but CCN was inhibited when treated with benazepril or sulodexide.Compared with the normal control group,thickened glomerular and tubular basement membrane,pyknosis,and increased collagen fiber in interstitium were found in the model group,but these pathological changes were lightened when treated with benazepril or sulodexide.Conclusion Sulodexide treatment could significantly reduce chronic cyclosporine nephropathy,and the effect is better than benazepril.
