摘要
以冬虫夏草与蛹虫草融合子菌丝体生物量为主要指标,采用正交设计方法对其菌丝体液体培养基进行筛选,比较3种培养方法对菌丝体的影响。结果表明其最适培养基配方为葡萄糖5.00g/L,蔗糖5.00g/L,酵母粉0.25g/L、黄豆粉5.00g/L(浸提液),KH2PO41.50g/L,MgSO4.7H2O 0.50g/L,VB14g/L,pH自然。25℃培养,周期156h,其菌丝体干重生物量可达3.51g/L。持续摇动培养法获得菌丝体生物量最多,利于菌体的规模发酵生产;动静相间法次之;静止法获得量较少。动静相间法所培养的菌丝体更有利于原生质体的制备和诱变育种。
Using mycelium biomass of fusant from Cordyceps sinensis and Cordyceps militaris as a major index, the liquid culture media of fusant mycelia was studied by means of orthogonal analysis. In order to study the growth of mycelia, three different cultural methods of the liquid culture were compared. The results showed that the optimal medium for the production of mycelia was glucose 5.00g/L, sucrose 5.00g/L, yeast powder 0.25g/L, soybean powder 5.00g/L, KH2PO4 1.50g/L, MgSO4TH2O 0.50g/L, VBI 4g/L, natural pH value. In the cultural condition of 25℃ and 168h, the mycelia biomass reached to 3.51g/L. When shaking cultivation conduced to a good deal of mycelia and stationary culture resulted in fewer mycelia. Swinging by hand intermittently conduced to the formation of protoplasts.
