摘要
目的通过小鼠血管通透性的变化,探讨地塞米松等药物缓解双硫仑样反应的作用机理。方法建立双硫仑样反应动物模型,每组16只昆明种小鼠分别给予地塞米松、维生素C、葡萄糖酸钙注射液干预处理,干预后以伊文思蓝(Evans blue,EB)法测微血管漏出率,同时取小鼠肌肉电镜观察不同干预组小鼠细胞结构变化。结果空白对照组细胞结构完整;双硫仑组细胞结构破坏明显;地塞米松组细胞质中小泡比双硫仑组少,但比对照组多;维生素C组细胞质中运输小泡减少;葡萄糖酸钙组胞质中运输小泡最多,胞质中线粒体肿胀。微血管通透性结果表明:双硫仑组EB漏出率最高,药物干预后漏出率降低。结论葡萄糖酸钙、维生素C和地塞米松,通过降低血管通透性减轻双硫仑样反应。
Objective To investigate the mechanism of clinical drugs including Dexamethasone on disulfiram-like reaction through the viewpoint of the vascular permeability of the rats. Methods The animal model of disulfiram-like reaction was established.Dexamethasone,vitamin C and calcium gluconate were infused into Kunming rats(16 cases in each group).The Evans blue(EB) leaking rate and tissue ultramicro pathologic change in electron microscope were detected. Results Tissue structure of the rats in electron microscope revealed that the cell structure was normal in the control group,while was obviously damaged in the disulfiram group.Dexamethasone treated group had roughly normal cell structure except more transport vesicles in cytoplasm than the control group.Vitamin C group had decreased transport vesicles but no swelling mitochondrion and no distending endoplasmic reticulum.Calcium gluconate group had the maximum transport vesicles and swelling mitochondrion but no distending endoplasmic reticulum.Tissue microvascular permeability results indicated that disulfiram group had the highest EB leaking rate,which was decreased after drug intervention except in the calcium gluconate group. Conclusion Calcium gluconate,vitamin C and Dexamethasone can eliminate disulfiram-like reaction by improving the vascular permeability.
