2型糖尿病患者脂肪组织中的HO-1表达及临床意义 被引量:2

Expression and significance of hemeoxygenase-1(HO-1) in subcutaneous and omental adipose tissue of type 2 diabetic patients

摘要 目的探讨2型糖尿病(T2DM)患者脂肪组织中血红素加氧酶-1(HO-1)基因及蛋白表达的临床意义。方法选择T2DM患者46例(T2DM组),其中伴肥胖者23例(T2DM+FAT组)、单纯T2DM 23例(单纯T2DM组);非T2DM患者56例(非T2DM组),其中单纯非T2DM患者33例(单纯非T2DM组)、伴肥胖者23例(非T2DM+FAT组)。采用RT-PCR技术检测各组腹部皮下及大网膜脂肪组织的HO-1 mRNA表达,蛋白免疫印迹技术检测其HO-1蛋白表达,分析其与临床及实验室相关指标的关系。结果①在腹部皮下及网膜脂肪组织中,单纯T2DM组及T2DM+FAT组的HO-1 mRNA表达量分别低于单纯非T2DM组及非T2DM组+FAT组,且T2DM+FAT组低于单纯T2DM组(P均<0.05)。网膜脂肪组织中的HO-1 mRNA表达量与BMI、HbA1c呈负相关(P均<0.01)。②在腹部皮下和网膜脂肪组织中,单纯T2DM组及T2DM+FAT组的HO-1蛋白表达量分别低于单纯非T2DM组及非T2DM组+FAT组(P均<0.05),其中T2DM+FAT组网膜脂肪组织的表达量与BMI、FPG、TG呈负相关(P均<0.05)。③相关分析显示,T2DM患者网膜脂肪组织HO-1蛋白表达量与血清脂联素水平呈正相关(P<0.05)。结论 T2DM患者的HO-1基因及蛋白表达明显降低,BMI越高,其表达量越低,HO-1表达降低可能与胰岛素抵抗及T2DM发生、发展有关。 Objective To observe the heme oxygenase-1 (HO-1) mRNA and protein expression in subcutaneous and omental adipose tissue of type 2 diabetic patients and their relation with serum adiponectin level, and other clinical indexes. Methods 23 type 2 diabetic patients with obesity, 23 type 2 diabetic patients, 33 controls with obesity and 23 controls were selected . The HO-1 mRNA expression level in adipose tissue was assayed by RT-PCR, and HO-1 protein expression level was assayed by Western blot. Being usedlinear correlation analysis, their correlations were observed. Results ①The expressions of HO-lmRNA in subcutaneous adipose tissue of type 2 diabetic patients with obesity were significantly lower (all P 〈0.05), and the level in omental adipose tissue was negatively correlated to BMI (P 〈0. 01 ). ② Type 2 diabetic patients and those with obesity showed significantly lower expressions of HO-1 protein in subcutaneous and omental adipose tissue comparing with control groups ( all P 〈 0.05 ), and the level in omental adipose tissue of type 2 diabetic patients with obesity were negatively correlated to BMI, FPG, TG ( all P 〈 0.05 ).③The expression level of HO-1 protein in omental adipose tissue of type 2 diabetic patients was positively correlated to the level of serum adiponectin ( P 〈 0.05 ). Conclusions The levels of HO-lmRNA and protein in type 2 diabetic patients are significantly lower than those of controls. The higher level of BMI is, the lower of HO-1 expression is. The low expression of HO-1 may be correlated with the occurrence and development of insulin resistance and DM.
