摘要
在单因素实验的基础上,采用正交实验法对麻疯树叶多酚提取时间,液料比,提取温度等提取条件进行优化,并采用1,1-二苯基-2-三硝基苯肼(DPPH)法,还原力法和β-胡萝卜素漂白法检测麻疯树叶中多酚类物质的抗氧化能力.试验确定麻疯树叶多酚类物质的提取最佳工艺为:40%乙醇,50℃,料液比1∶40,提取80min.麻疯树叶多酚的抗氧化能力较强,其清除DPPH自由基的EC50为0.15mg/mL,还原力测定的EC50为2.06mg/mL,β-胡萝卜素/亚油酸体系中测定BHT和麻疯树叶提取物都有抑制作用.麻疯树叶多酚类物质具较好的抗氧化作用.
Based on single-factor experiment results,four factors were analyzed with response Orthogona test,including effects of extraction time,ratio of solvent to material,extraction temperature and ethanol concentration.The antioxidant activity of Jatropha curcas L was evaluated with DPPH(1,1-diphenyl-2-picryl-hydrazyl) method,ferric reducing/antioxidant power(FRAP) and β-carotene bleaching assay.The optimum condition for extraction of polyphenols from Jatropha curcas L.with ethanol solution was ethanol concentration of 40%,extraction temperature of 50 ℃,extraction time of 80 min and material-liquid ratio of 1∶40.The EC50 values for DPPH,and ferric reducing/antioxidant power(FRAP) were 0.15 and 2.06 mg/mL for the ethanol-based extract.In the β-carotene-linoleatesy system,the BHT and ethanol concentration both showed inhibition.The polyphenol of Jatropha curcas L.possesses antioxidant activity.
