摘要
目的探讨男性精浆中细胞因子IL-2、IL-6、TNF-α浓度和氧化应激指标活性氧、丙二醛水平的关系。方法筛选41例参数正常男性、65例少弱精子症及21例无精子症患者精液,离心后分离精浆,ELISA方法测定不同组细胞因子IL-2、IL-6、TNF-α浓度,化学发光法测定精液活性氧(ROS)水平,生化比色法测定精浆中的丙二醛(MDA)浓度,并分析细胞因子浓度与氧化应激指标活性氧、丙二醛水平变化的相关性。结果 IL-2在少弱精子症组精浆中最高,与其余两组相比差异有显著性,而在精液参数正常组与无精子症组间无明显差异;IL-6、TNF-α在少弱精子症与无精子症组精浆中浓度均较精液参数正常组高,差异有显著性,而在少弱精子症与无精子症组之间差异无显著性;精浆中ROS与MDA在少弱精子症组最高,与另两组相比差异有显著性,而无精子症组与精液参数正常组间差异无显著性。在不同组精浆中ROS、MDA水平变化与IL-2浓度变化相同,而精浆中IL-6和TNF-α的变化趋势与氧化应激指标的变化无关。结论精浆中细胞因子水平变化与男性不育有关,质量异常的精液中细胞因子浓度明显升高,而少弱精子症导致的男性生育力降低与氧化应激有关。
Objective To explore the relationship of level of IL-2, IL-6, TNF-α, with reactive oxygen species (ROS) and malondialdehyde in different seminal plasma. Methods 41 male patients with normal fertility, 65 patients with oligospermia and asthenospermia and 21 patients with azoospermia were enrolled. After the removal of seminal by centrifugation, levels of IL-2, IL-6, TNF-α were detected by ELISA, level of ROS by chemiluminescence and level of MDA by biochemical colorimetric method. The concerntration of cytokines above and oxidative stress related index was analysed. Results The level of IL-2 was highest in patients with oligospermia and asthenospermia, but no difference was detected in patients with normal fertility and azoospermia. Levels of IL-6, TNF-α were higher in patients with oligospermia, asthenospermia and asthenospermia than in normal male, but no difference was detected in patients with oligospermia, asthenospermia and asthenospermia. Levels of ROS and MDA were highest in patients with oligospermia and asthenospermia, but no difference was detected in patients with normal fertility and azoospermia. Level of ROS and MDA was correlated with level of IL-2 in different semen. Conclusions Seminal cytokines was closely related with male infertility. The level of cytokines increased obviously in abnormal semen, and low male reproduction resulted from oligospermia and asthenospermia is correlated to oxidative stress.
