腺苷A3受体激动剂对新生期母婴分离肠易激综合征大鼠内脏高敏感的作用

摘要 目的研究腺苷A3受体在新生期母婴分离肠易激综合征(IBS)大鼠模型内脏高敏感中的作用。方法采用新生期Sprague-Dawley雄性大鼠和母婴分离方法建立内脏高敏感模型,分别用腺苷A3受体激动剂2-Cl-IB-MECA及DMSO溶剂作用后,用行为学观察和腹部回缩反射评分来检测内脏痛反应,用免疫组化方法检测腰骶段脊髓背角神经元c-fos蛋白的表达数量和远端结肠中5-羟色胺阳性细胞数目,用ELISA方法检测远端结肠中5-羟色胺的含量。结肠行常规组织学检测。结果与DMSO组比较,2-Cl-IB-MECA组大鼠在40、60和80 mmHg结直肠压力扩张下的AWR评分显著减低(P<0.05),脊髓背角c-fos蛋白免疫反应阳性神经元数目显著减少(P<0.05),结肠黏膜5-羟色胺阳性细胞数目和5-羟色胺含量均显著减少(P<0.05)。两组大鼠在结直肠压力扩张前后,结肠均未见组织损伤或炎症。结论腺苷A3受体激活可能通过减少结肠5-羟色胺的作用未减轻IBS大鼠模型的内脏高敏感,腺苷A3受体可能是治疗IBS内脏高敏感的新靶点。
