摘要
目的探索活跃链霉菌36#摇瓶发酵合成那西肽的固体斜面培养条件的最佳工艺。方法利用单因素与正交试验对固体斜面培养条件进行优化。结果得到合成那西肽的活跃链霉菌36#的固体斜面最优培养条件为:初始phi7.8,培养温度为37℃,培养时间为3d,冷藏时间为O一3周,那西肽产量可达936mg/L。与原工艺相比较,那西肽的产量增加了13.8%。结论通过活跃链霉菌固体斜面培养条件的优化,可以提高活跃链霉菌摇瓶发酵合成那西肽的产量。
Objective The slant cultivation conditions of Streptomyces actuosus 36# were investigated to explore optimum process of fermentation synthesis of nosiheptide in shake flask. Methods Single factors experiment and orthogonal design were used to optimize the conditions of the slant culture. Results The highest production of nosiheptide (936mg/L) was obtained under the following slant cultivation conditions: initial pH 7.8, culture temperature 37℃, incubation time 3d and cold store time 0-3w, which was increased by 13.8% compared to the original process. Conclusion The high yield of nosihcptide in shake flask could be obtained by optimizing the slant cultivation conditions of Streptomyces actuosus.
