摘要
以乙醇为提取溶剂,回流提取新疆昆仑雪菊原花青素。在单因素实验的基础上,根据Box-Behnken的中心组合实验设计原理,运用Minitab15数据统计分析软件,采用4因素3水平的响应面分析法,以昆仑雪菊原花青素提取得率为响应值,研究了乙醇浓度、料液比、浸提时间和浸提温度对昆仑雪菊原花青素提取得率的影响,并优化了提取工艺。确定了回流提取昆仑雪菊原花青素的最佳工艺条件为:乙醇浓度60%、料液比为1:16、浸提时间为2.5h、浸提温度为60℃,在此条件下昆仑雪菊原花青素的提取率达到10.24%。
Procyanidins from Kunlun Chrysanthemum in Xinjiang was obstained by reflux extraction method with alcohol as extraction solvent.The preparation process of procyanidins from Kunlun Chrysanthemum was optimized according to Box-Behnken central composition design principle on foundation of single factor experiment,the method of response surface methodology(RSM) with 4 factors and 3 levels combined with Minitab15-statistical data analysis software was adopted with the extraction yield of procyanidins from Kunlun Chrysanthemum as the response value.The effect of ethanol concentration,solid-liquid ratio,extraction time and extraction temperature on the efficiency of extraction of procyanidins from Kunlun Chrysanthemum were studied in this paper.According to the above results,the optimum preparation conditions were obtained as follows:ethanol concentration 60%,ratio of material to liquid 1:16,extraction time 2.5 h,and extraction temperature 60 ℃.Under these conditions,the extracting ratio of procyanidins from Kunlun Chrysanthemum reached 10.24%.
