摘要
根据已发表的禽脑脊髓炎病毒的VP2基因序列,设计并合成了6对特异扩增禽脑脊髓炎病毒VP2基因片段的引物,通过条件优化,成功建立了针对禽脑脊髓炎病毒的环介导等温扩增(LAMP)检测法,并测定其特异性和敏感性,对采集的禽临床样品的核酸分别进行了检测。结果表明,该法只检出禽脑脊髓炎病毒。敏感性扩增结果表明,Lamp检测禽脑脊髓炎病毒为100fg的RNA模板。该LAMP方法有简便、快速和特异性高的优点,可用于临床上对禽脑脊髓炎病毒的快速检测。
A set of six specific oligonucleotide primers was designed and synthesized to recognize six distinct genomic sequences of the VP2 from AEV in GenBank. The LAMP method was established to detect AE through the optimized reaction conditions. Then,the pathogenic bacteria and clinical samples from chicken were detected with this LAMP. The results showed that the LAMP assay detected all 5 reference and isolated AE strains, did not cross-react with other avian pathogens. The limit of detection was 100 fg of RNA template of AE. Therefore, this LAMP assay should be a good alternative method for specific diagnosis of AE from clinical samples.
