GnRH在黑线仓鼠不同器官中的差异表达

摘要 该研究以野生黑线仓鼠为材料,用实时定量PCR方法检测GnRH在下丘脑和部分生殖器官(睾丸、附睾、储精囊、卵巢、子宫)中的表达量,并分析个体间的表达差异.研究结果表明:①GnRH在黑线仓鼠的下丘脑、睾丸、附睾、储精囊、卵巢、子宫中均有表达,其中在下丘脑中的表达量相对较高而在生殖器官则表达量较低;②在雄性个体中,GnRH在下丘脑、睾丸、附睾、储精囊四种组织中表达量的比值为1∶0.166∶0.189∶0.073;③在雌性个体中,GnRH在下丘脑、卵巢.子宫三种组织中表达量的比值为1∶0.304∶0.040.研究结果显示,GnRH在不同组织和不同个体间均存在差异性表达,提示GnRH既可以通过下丘脑直接影响生殖功能,又可以直接参与生殖系统功能的调控,但其调控机制可能有所不同.研究结果将为研究GnRH在黑线仓鼠的功能及分子调控机制奠定基础,为探究野生黑线仓鼠繁殖动态的遗传调控机制提供理论保障,并为农田鼠害的综合防治提供科学依据. Hypothalamus and part of the reproductive organs (testis, epididymis, seminal vesicle, ovary, ute- rus) of Cricetulus barabensis were chosen as materials in this study to detect the differentiated expression of GnRH by real-time PCR and analysised the differences among individuals. The results proved that ① GnRH expresses in hypothalamus, testis, epididymis, seminal vesicle, ovary and uterus. The value of GnRH express in hypothalamus is high, while the value in other organs were extremely low. ② In the male, the ratio of the expression value in hy- pothalamus, testis, epididymis, seminal vesicle is 1: 0. 166: 0. 189: 0. 073. ③ In the female, the ratio of the ex- pression value in hypothalamus, ovary and uterus is 1: 0. 304: 0. 040. The results show that there is expression di- versity of GnRH mRNA in different tissues as well as different individuals, also GnRH has significant affect on the whole reproductive system and the regulating system of hypothalamus, but ks regulatory mechanism may be differ- ent. The results will provide references for the study on molecular mechanism of GnRH and a theoretical basis to explore the genetic regulatory mechanism of propagation dynamics in Cricetulus barabensis
