组蛋白甲基化修饰对DNA损伤的影响及其与肿瘤的关系

摘要 细胞内部因素或周围环境因素会导致DNA损伤,DNA损伤可触发两条途径来维持基因组的稳定性:(1)细胞周期阻滞,由DNA损伤修复蛋白介导DNA损伤修复;(2)严重的DNA损伤会由p53介导进入细胞凋亡途径。两者发生异常时,损伤的DNA累积而导致基因突变,最严重的后果是癌变。研究发现组蛋白甲基化修饰在DNA损伤反应过程中可能发挥重要的调节作用,而组蛋白去甲基化酶的相继发现,证明了这一修饰过程的可逆性。此文主要对DNA损伤与肿瘤的关系,以及组蛋白甲基化修饰在DNA损伤反应过程中的调节作用进行综述。
