甘蔗茎全长cDNA文库构建及EST序列分析 被引量:2

Construction of Full-length cDNA Library from Sugarcane Stem and the Corresponding EST Analysis

摘要 在利用改进的Oligo-capping法构建1个高质量的甘蔗茎全长cDNA文库基础上,对该文库进行大规模测序,获得了283条5′端有效EST序列,平均长度为459 bp,经聚类和拼接获得204个假定独立转录本(TUTs),冗余序列所占比例27.9%。NCBI同源比对分析结果表明,其中132条EST拼接的103个TUTs与已知功能基因具有较高的同源性,结合拟南芥功能基因分类标准对这些TUTs进行分类,可分为12类,以参与蛋白质合成的基因最多,其次为细胞结构、蛋白质降解和贮藏、转录等类型的基因。 Sugarcane is not only the most important sugar plant, but also one of the most important energy plants in the world. Based on the sugarcane stem full-length cDNA library successfully constructed by modified Oligo-capping, we obtained 283 ESTs (expressed sequence tags) of 5'end from this cDNA library by high-throughput sequencing, the average length of these ESTs was 459 bp, and 204 TUTs (tentative unique transcript, TUT) were obtained after clustering and splicing. The rate of the redundant sequence of the library was 27.9%. NCBI homologies analysis showed that these 103 TUTs (including 132 ESTs) were determined to the tags of gene with putative function, and were classified intol2 categories according to the functional classification of Arabidopsis genes, among which the largest category was genes related to protein synthesis, the others were cell structure, protein destination and storage, transcription etc.
