补肾活血法对阿霉素肾病小鼠肾组织wt1表达的影响 被引量:2

Effect of nourishing kidney and activating blood recipe of traditional Chinese medicine on expression of wt1 in renal tissues of mice with adriamycin induced nephropathy

摘要 目的观察补肾活血法中药对阿霉素诱导的阿霉素肾病小鼠肾组织wt1表达水平的影响。方法雄性BALB/c小鼠24只随机分为对照组、阿霉素肾病模型组和补肾活血中药治疗组,采用尾静脉注射阿霉素法建立肾病模型,收集各组血、尿及肾组织,检测血浆白蛋白(ALB)、体质量、右肾质量及尿蛋白排泄率(UAE);光学显微镜下观察肾组织形态改变;间接免疫荧光法检测各组肾组织wt1的表达;反转录-聚合酶链反应(RT-PCR)法检测各组肾组织wt1mRNA水平。结果肾病模型组ALB([20.160±0.626)g/L]明显低于对照组([26.728±0.518)g/L]与治疗组([26.167±0.594)g/L],肾病模型组UAE([70.453±0.603)mg/24h]明显高于对照组([11.499±0.146)mg/24h]和治疗组([30.784±0.526)mg/24h],差异均有统计学意义(P<0.05)。对照组肾小球wt1呈阳性表达,模型组表达明显减弱,治疗组较模型组阳性表达增强。对照组和治疗组wt1mRNA表达水平均高于模型组,差异有统计学意义(P<0.01),对照组和治疗组的wt1mRNA表达水平差异无统计学意义(P>0.05)。结论补肾活血法药物可以通过上调wt1的表达维持足细胞的正常功能,从而减轻小鼠的肾脏损伤。 Objective To investigate the effects of nourishing kidney and activating blood recipe of traditional Chinese medicine on the expression of wtl in renal tissues of mice with adriamycin-induced nephropathy. Methods Twenty-four male BALB/c mice were randomly divided into three groups, control group, model group (adriamycin-induced nephropathy ) and treatment group with nourishing kidney and activating blood recipe of traditional Chinese medicine. The mouse model was prepared by caudal vein injection of adriamycin. Blood, urine and renal tissues were collected from three groups. The serum albumin (ALB), body weight, right renal weight and urine album excretion (UAE) were detected. Pathological change of renal tissues was examined under light microscope. The wtl protein expression levels in renal tissues were determined by immunofluorescence assay. The expression levels of mRNA in renal tissues were measured by RT-PCR assay. Results ALB in model group was (20.160 _± 0.626)g/L, which was significantly lower than that in control group [(26.728±0.518)g/L] and treatment group [(26.167±0.594)g/L] (P〈0.05). UAE in model group was (70.453±0.603)mg/24 h, which was significantly higher than that in control group [( 11.499±0.146)mg/24 h] and treatmeat group [(30.784±0.526)mg/24 h]. The wtl protein expression level and wtl mRNA expression level of renal tissues in model group was significantly lower than those in control and treatment groups(P〈0.01 ). There was no significant difference of wtl mRNA expression level between treatment group and control group. Conclusion The treatment with nourishing kidney and activating blood recipe of traditional Chinese medicine could up-regulate the expression of wtl to maintain the normal cellular function and relieve renal injury.
