摘要
目的探讨尿液特异基因DD3检测在前列腺癌(prostate cancer,PCa)诊断中的应用价值。方法选择血清前列腺特异性抗原(PSA)异常患者80例,均行前列腺穿刺活检,诊断为前列腺癌27例,前列腺增生53例。留取患者前列腺按摩后尿液。另留取20例健康者尿液作为正常对照。使用环介导等温扩增技术(loop-mediated isothermal amplification,LAMP)检测受试者尿液中DD3表达,并将结果与前列腺穿刺活检结果进行对比。结果前列腺癌患者尿液DD3均阳性表达,前列腺增生患者中仅1例DD3阳性表达,正常对照组DD3均阴性。结论 LAMP法检测尿液DD3与病理检测结果具有较高的一致性,且方法简单、快捷,对前列腺癌诊断具有重要的价值。
Objective To explore the application value in diagnosis of prostate cancer by testing the specific DD3 mRNA in urine. Methods A total of 80 patients with abnormal prostate specific antigen (PSA) in blood serum underwent prostate puncture biopsy, and 27 patients were diagnosed as having PCa, and 53 patients were diagnosed as having benign prostatic hyperplasia (BPH). Patients'urine samples were collected after prostate chirapsia. In addition, 20 urine samples from healthy people were collected as the control group. DD3 expressions in urine from subjects were detected with loop-mediated isothermal amplification (LAMP) technique, and the results were compared with that of prostate biopsy. Results The DD3 examinations revealed positive expression in all urine samples from patients with PCa, a urine sample from patient with BPH, and in control group, urine samples were all negative expressions. Conclusion The results by LAMP in detection of DD3 in urine samples are highly consistent with the results of prostate biopsy. This method is important for diagnosis of prostatc cancer.
