HPLC法测定咳嗽枇杷糖浆中京尼平苷酸和毛蕊花糖苷

Determination of geniposidic acid and verbascoside in kesoupipa syrup by HPLC

目的用高效液相色谱梯度洗脱法同时测定咳嗽枇杷糖浆(枇杷叶、桔梗、麻黄、车前子等)中京尼平苷酸和毛蕊花糖苷,为制定该制剂质量标准中定量测定方法及限度提供依据。方法色谱柱:Agilent-C18(4.6 mm×150 mm,5μm)流动相:以甲醇-0.5%醋酸溶液梯度洗脱(0～1 min:甲醇质量分数为5%;1～40 min:甲醇质量分数由5%递升至60%;40～50 min:甲醇质量分数由60%递减至5%);流速:1.0 mL.min-1,检测波长:254 nm,柱温:30℃,进样量:10μL。结果京尼平苷酸和毛蕊花糖苷的线性范围为0.8128～4.064μg和0.7696～3.848μg,平均加样回收率(n=6)分别为99.66%和99.14%。结论该方法简单、快速、准确,可作为咳嗽枇杷糖浆的质量控制方法。

Objective To establish a method for determing in geniposidic acid and verbascoside in kesoupipa syrup（eriobotryae folium, platycodonis radix, ephedrae herba, plantaginis semen, etc）. Methods The HPLC method was carried out on Agilent C ls column （4.6 mm ×150 mm,5μm）. The mobile phase was methanol-015 % acetic acid （ 0- 1 min ：5： 95 ; 1-40 min ：60： 40 ; 40-50 min ： 5： 95 ）. The flow rate was 1.0 mL·min^-1. The detection wavelength was at 254 nm,The column temperature was set at 30℃ with 10 μL injection volumn. Results The linear range of geniposidic acid was 0.8128 - 4. 064 μg, and that of verbaseoside was 0.7696- 3. 848μg. Their average recoveries were 99.66% and 99.14%, respectively. Conclusion The method is accurate and can be used for the quality control of kesoupipa syrup.
