摘要
目的探讨不同浓度医用三氧对离体肝癌细胞SMMC-7721凋亡率的影响。方法用医用三氧发生器生成3种不同浓度(20,50和80μg/ml)的三氧,分别将其通入瓶装三蒸水100ml 10min,生成不同浓度的三氧水。实验分为5组,A组为0μg/ml03,B组为20μg/mlO3,C组为50μg/mlO3,D组为80μg/mlO3,E组为对照组。将肝癌细胞SMMC-7721接种于6孔培养板,分别加入不同浓度的三氧水培养32h后消化、收集细胞到10m1的离心管中,采用AnnexinV—FITC/PI双染法染色,然后用流式细胞仪检测细胞的凋亡率。结果B、C、D组均出现SMMC-7721细胞凋亡;各组与D组比较差异均有统计学意义(P〈0.05)。结论3种不同浓度三氧化水均可导致SMMC-7721细胞凋亡,以80μg/mlO3出现的凋亡率最高。
Objective To explore the effects of ozone in different concentration on the apopto- sis rate of liver cancer SMMC-7721 in vitro. Methods Theraputic ozonation water was produced by the ozone generator and went through 100 ml bottled medical tri-distilled water for 10 min in 3 differ- ent concentration (20,50 and 80μg/ml). There were 5 groups in this experiment. Ozone was 0 μg/ml in group A, and 20μg/ml,50 μg/ml,80 μg/ml 03 in group B,C,D respectively and E was control group. Liver cancer SMMC-7721 cells were planted in a 6 wells plates. The cells were collected into the 10 ml centrifuge tubes after 32 h with ozonation water and then dyeing with the annexin V-FITC. The apoptosis rate was detected by the flow cytometry (FCM). Results Cells in group B,group C and group D showed apoptosis. There were significant differences between group D and other groups (P〈0.05). Conclusion Ozonation water can induce SMMC-7721 apoptosis in 3 different concentra- tions (20, 50 and 80 μg/ml). The highest apoptosis rate is in group D with ozone concentr- ation 80μg/ml.
