摘要
目的探讨大黄素对IL-1β诱导下的血管平滑肌细胞迁移以及VCAM-1、MCP-1表达的影响。方法将10ng/ml的IL-1β作用于血管平滑肌细胞,加入10μmol/L大黄素进行干预,实验设置对照组、IL-1β组、IL-1β+大黄素组。Transwell法检测各组细胞的迁移能力;RT—PCR和ELISA检测各组细胞MMP-2、MMP-9的表达;RT-PCR和Western检测各组细胞VCAM-1和MCP-1的表达。结果IL-1β组细胞迁移能力显著高于对照组(P〈0.05),IL-1β+大黄素组细胞迁移能力显著低于IL-1β组(P〈0.05);IL-1β组MMP-2、MMP-9、VCAM-1、MCP-1mRNA以及蛋白的表达均显著高于对照组(P〈0.05),与IL-1β组相比IL-1β+大黄素组MMP-2、MMP-9、VCAM-1、MCP-1mRNA以及蛋白的表达均显著降低(P〈0.05)。结论大黄素能够有效抑制IL-1β诱导的血管平滑肌细胞迁移并降低其VCAM-1、MCP-1的表达。
Objective To investigate the effects of emodin on migration and VCAM-1, MCP-1 expression of IL-1β induced vascular smooth muscle cells (VSMCs) . Methods VSMCs was cul- tured and incubated with IL-1β (10 ng/ml) , emodin (10 p, mol/L) was added and migration of VSMCs is detected by transwell assay, expression of MMP-2, MMP-9, VCAM-1 and MCP-1 is de- tected by RT-PCR, ELISA or Western blot. Results The migration of IL-1β group is increased sig- nificantly ( P 〈 0. 05), but decreased significantly in IL-1β + emodin group when compared to IL-1β group (P 〈 0. 05) . mRNA and protein level of MMP-2, MMP-9, VCAM-1 and MCP-1 is increased significantly in IL-1β group ( P 〈 0. 05 ), and decreased significantly in IL-1β + emodin group when compared to IL-1 (3 group (P 〈 0. 05 ) . Conclusion Emodin can effectively inhibits the migration and the expression of VCAM-1, MCP-1 in VSMCs which induced by IL-1β.
