摘要
目的探讨人绒毛膜促性腺激素(humanchorionicgonadotropin,hCG)对乳腺癌细胞侵袭能力的作用。方法应用MTT法、体外侵袭转移实验方法、半定量RT—PCR,观察hCG对高表达LHR(1uteinizinghormone/humanchorionicgonadotropinreceptor)的MCF.7乳腺癌细胞生长、侵袭能力和mmpl、romp2、mmp9、timpl、timp3、vegf等侵袭转移相关基因转录表达的影响。结果hCG对高表达LHR的MCF一7乳腺癌细胞增殖力无明显影响,对侵袭力有明显抑制作用,对romp9、veff基因转录起抑制作用,对timpl基因转录起促进作用。结论hCG对高表达LHR的MCF一7乳腺癌细胞侵袭能力起抑制作用,对mmp9、red基因表达起抑制作用;对timpl基因表达起促进作用。
Objective To investigate the effect of human chorionic gonadotropin (hCG) on the invasion of breast cancer cells. Methods MCF-7 cells, a breast cancer cell line over-expressing LHR, were cultured for the study. The effect of hCG on the growth and invasion of MCF-7 cells was examined by using MTF method and in vitro invasion and metastasis experimental methods. The ex- pression of invasion-and metastasis-related genes including mmpl, romp2, romp9, timpl, timp3 and vegf was detected in MCF-7 cells by semi-quantitative RT-PCR. Results The proliferative abil- ity of MCF-7 cells was not obviously influenced by hCG, while the invasive ability was significantly inhibited. The transcription of romp9 and vegf genes was suppressed, but the timpl gene transcrip- tion was enhanced in MCF-7 cells. Conclusion hCG may suppress the invasion of breast cancer cells, which may involve a decrease in the transcription of romp9 and vegf genes and an increase in the timpl gene transcription.
